Selection System for Molecular Biological Studies: clonNAT + Plasmid pYL16

clonNAT [CAS 96736-11-7] is the trade name of the natural product nourseothricin (a complex of the streptothricins F, E and D), produced by Streptomyces noursei, isolated as dihydrogen sulphates and inhibits bacteria, mycobacteria, mycoplasms, protozoae, yeasts, viruses and plants. The mechanism of action involves in the inhibition of ribosomal protein synthesis and induction of miscoding. Plasmid pYL16 and clonNAT can be used as a selection system for microbiological applications and for modern plant cultivation. Plasmid pYL16 contains the resistance gene nat1 from Streptomyces noursei. The corresponding resistance genes sat1, sat2, sat3 come from Escherichia coli, stat from Streptomyces lavendulae, encoding a nourseothricin N-acetyltransferase. The mechanism of developing resistance in all cases is the monoacetylation of the b-amino group of the b-lysyl moiety of clonNAT. Plasmid pYL16 is smaller than the plasmids pINS1 and pHN15 used earlier. The resistance genes are available as markers for heterologous expressions.clonNAT may be used in fermentations of recombinant clones to maintain the permanent propagation of vectors carrying one of the resistance genes.

Summarizing some advantages of the selection system clonNAT + plasmid pYL16 in comparison with commercially available selection systems, it can be said that:

clonNAT

  • is not used as a drug in human medicine,
  • possesses a broad activity spectrum,
  • needs only low selective concentrations,
  • shows no cross resistance with therapeutic antibiotics and selective agents in other systems,
  • keeps the intracellular resistance determinants in the medium actively,
  • is exceptionally useful for selection of recombinant yeast strains (no cross reactivity with aminoglycosides G418 and hygromycin B, low or zero background, small gene
    [A. Goldstein, 1999]),
